Generation of highly productive recombinant cell lines for biomanufacturing is a labor-intensive and time-consuming process. DNA silencing has been shown to hinder recombinant gene expression following transfection and chromosomal integration. The Ubiquitous Chromatin Opening Element (UCOE®) technology enables linked gene expression cassettes to overcome DNA methylation-mediated silencing, thus reducing the time and resources required to generate highly productive recombinant cell lines. We will describe the mode of action of the UCOE® elements, discuss how to integrate the UCOE® technology into cell line development platforms, and provide data using the UCOE® technology for cell line development in an industry-relevant context.

Dr. Michael Antoniou established his own research group at King’s College London in 1994. His research interests have centered on identifying and characterizing genetic regulatory elements with a dominant chromatin remodeling and transcriptional activating capability. In collaboration with industrial partners he discovered ubiquitous chromatin opening elements (UCOEs) during the 1990’s and has since been extensively involved in developing their biotechnological applications in both a biomanufacturing and especially human gene therapy vector context. Dr. Antoniou holds inventor status on a number of patents covering UCOE® gene expression technology.

Dr. Joe Orlando joined EMD Millipore in 2007 and has since led various technology projects to support the Bioprocess industry. As a member of the Process Solutions, he has instituted Ubiquitous Chromatin Opening Element (UCOE) technology at the center of EMD Millipore’s biomanufacturing recombinant protein expression platform. Further, he has worked with other groups in the organization to complete the USP and DSP technologies to establish a complete platform.

Dr. Orlando received his Ph.D. in Microbiology and Immunology from Wake Forrest University in Winston-Salem, North Carolina and completed his postdoctoral fellowship at Harvard Medical School in Boston, MA, USA. He has published over 10 articles and patents.
